| Abstract: | Prevalence of faecal coliform bacteria and the survival of Escherichia coli, Vibrio parahaemolyticus and Salmonella paratyphi were studied in the water and sediment from Vembanadu Lake in the presence and absence of protozoan predators. The density of faecal coliform bacteria ranged between mean MPN value 5080–9000/100 ml in water and 110,000–988,000/1 g in sediment (p <0.01), which was 110 times greater than in overlying water. The laboratory microcosm studies revealed that E. coli, V. parahaemolyticus and S. paratyphi showed significantly higher survival (p <0.05) potential in sediment than in overlying water both in the presence and absence of protozoan predators. The results indicate that Vembanadu Lake sediment constitutes a reservoir of pathogenic bacteria and exhibits potential health hazard from possible resuspension and subsequent ingestion during recreational activities. Therefore, assessment of bacterial concentration in freshwater lake sediments used for contact and non-contact recreation is of considerable significance for the proper assessment of microbial pollution of the overlying water and the management and protection of related health risk at specific recreational sites. In addition, assessment of the bacterial concentration in sediments can be used as a relatively stable indicator of long-term mean bacterial concentration in the water column above. |

| Abstract: | Microcosm studies were performed to evaluate the survival of Escherichia coli, Salmonella paratyphi and Vibrio parahaemolyticus in water and sediment collected from the freshwater region of Vembanad Lake (9 35◦N 76 25◦E) along the south west coast of India. All three test microorganisms showed significantly (p < 0.01) higher survival in sediment compared to overlying water. The survival in different sediment types with different particle size and organic carbon content revealed that sediment with small particle size and high organic carbon content could enhance their extended survival (p < 0.05). | Abstract: | Broiler chicken is gaining popularity among the consumers of India. Since poultry is recognised as a leading food vehicle for Salmonella contamination, the prevalence and distribution of Salmonella serotypes in broiler chickens and processing environments of retail outlets has been studied. In the present study 214 samples of broiler chicken and 311 environmental samples from cage were analysed for the presence of Salmonella. Of the various body parts of live chicken analysed prevalence varied from 1.4% in cloacca to 6.9% in crop region. Environmental samples from the cage showed higher prevalence of Salmonella ranging from0 to 16.67%. Apart from Salmonella enteritidis, which was the predominant Salmonella serotype in the chickens as well as in the environmental samples, other serotypes such as S. bareilly, S. cerro, S. mbandaka and S. moladewere also encountered. | Abstract: | To assess the prevalence of faecal coliform bacteria and multiple drug resistance among Escherichia coli and Salmonella serotypes from Vembanadu Lake. Study design: Systematic microbiological testing. Methods: Monthly collection of water samples were made from ten stations on the southern and northern parts of a salt water regulator constructed in Vembanadu Lake in order to prevent incursion of seawater during certain periods of the year. Density of faecal colifrom bacteria was estimated. E. coli and Salmonella were isolated and their different serotypes were identified. Antibiotic resistance analysis of E. coli and Salmonella serotypes was done and the MAR index of individual isolates was calculated. Results: Density of faecal coliform bacteria ranged from mean MPN value 2900 -7100/100ml. Results showed multiple drug resistance pattern among the bacterial isolates. E. coli showed more than 50% resistance to amickacin, oxytetracycline, streptomycin, tetracycline and kanamycin while Salmonella showed high resistance to oxytetracycline, streptomycin, tetracycline and ampicillin. The MAR indexing of the isolates showed that they have originated from high risk source such as humans, poultry and dairy cows. Conclusions: The high density of faecal coliform bacteria and prevalence of multi drug resistant E. coli and Salmonella serotypes in the lake may pose severe public health risk through related water borne and food borne outbreaks |

| Abstract: | A yearlong (September 2009–August 2010) study was undertaken to find out possible reasons for occasional occurrence of White Spot Syndrome Virus (WSSV) outbreak in the traditional prawn farms adjoining Cochin backwaters. Physicochemical and bacteriological parameters of water and sediment from feeder canal and four shrimp farms were monitored on a fortnightly basis. The physicochemical parameters showed variation during the two production cycles and between the farms studied. Dissolved oxygen (DO) content of water fromfeeder canal showed low oxygen levels (as low as 0.8mg/L) throughout the study period. There was no disease outbreak in the perennial ponds. Poor water exchange coupled with nutrient loading from adjacent houses resulted in phytoplankton bloom in shallow seasonal ponds which led to hypoxic conditions in early morning and supersaturation of DO in the afternoon besides considerably high alkaline pH. Ammonia levels were found to be very high in these ponds.WSSV outbreak was encountered twice during the study leading to mass mortalities in the seasonal ponds. The hypoxia and high ammonia content in water and abrupt fluctuations in temperature, salinity and pH might lead to considerable stress in the shrimps triggeringWSSV infection in these traditional ponds |

| Abstract: | TThe invention of novel antibiotics and other bioactive microbial metabolites continues to be an important aim in new drug discovery programmes. Actinomycetes have the potential to synthesize lots of diverse biologically vigorous secondary metabolites and in the last decades actinomycetes became the most productive source for antibiotics. Therefore in the present study we analyze the antibacterial activity of the actinomycetes isolated from grassland soil samples of Tropical Montane forest. A total of 33 actinomycete strains isolated were characterized and screened for antibacterial activities using well diffusion method against six specific pathogenic organisms. Identification of the isolates revealed that the majority of them were belonging to Streptomycetes followed by Nocardia, Micromonospora, Pseudonocardia, Streptosporangium, Nocardiopsis and Saccharomonospora. Among the 33 isolates, Gr1 strain showed antagonistic activity against all checked pathogens. Nine strains showed antibacaterial activity against Listeria, Vibrio cholera, Bacillus cereus, Staphylococcus aureus and Salmonella typhi and only 2 strains (Gr1and Gr25) showed antagonism to E. coli. The overall percentage of activity of actinomycetes isolates against each pathogenic bacterium was also calculated. While 63.63% of the actinomycetes were antagoinistic against Listeria, Vibrio cholerae, and Bacillus cereus, 60.6% of them were antagonistic to Staphylococcus aureus. Very few isolates (6.06%) showed antibacterial activity against E. coli. In general most of the actinomycetes isolates were antagonistic to grampositive bacteria such as Listeria, Bacillus and Staphylococcus than Gram-negative bacteria Vibrio cholerae, E. coli and Salmonella |

| Abstract: | The objective of the present study was to assess the prevalence of various motile aeromonads in freshwater ornamental fishes and to elucidate the antibiogram and beta hemolytic activity among the isolates. A total of 120 ornamental fish samples were screened and analyzed for Aeromonas spp. Motile aeromonads were isolated from 37.5% of the ornamental fish samples. Various species of motile aeromonads such as Aeromonas caviae, Aeromonas hydrophila, Aeromonas jandaei, Aeromonas schubertii, Aeromonas sobria, Aeromonas trota and Aeromonas veronii were detected. All the isolates were sensitive to ceftazidime, chloramphenicol, ciprofloxacin and gentamicin. Multiple antibiotic resistance was observed in 58% of the isolates. |

| Abstract: | Actinomycetes are gram-positive, free-living, saprophytic bacteria widely distributed in soil, water and colonizing plants showing marked chemical and morphological diversity. They are potential source of many bioactive compounds, which have diverse clinical effects and important applications in human medicine. In the present work, we have studied some of the physiological and biochemical characteristics of 36 actinomycete strains isolated from the shola soils of tropical montane forest; a relatively unexplored biodiversity hotspot. Ability of actinomycetes isolates to ferment and produce acids from various carbohydrate sources such as innositol, mannose, sorbitol, galactose, mannitol, xylose, rhamnose, arabinose, lactose and fructose were studied. Almost all the carbon compounds were utilized by one or other actinomycete isolates. The most preferred carbon sources were found to be xylose (94.44%) followed by fructose and mannose (91.66%). Only 41.76% of the isolates were able to ferment lactose. The ability of actinomycetes isolates to decompose protein and amino acid differ considerably. 72.22% of the isolates were able to decompose milk protein casein and 61.11% of the isolates decompose tyrosine. Only 8.33% of the strains were able to decompose amino acid hypoxanthine and none of them were able to decompose amino acid xanthine. Potential of the actinomycetes isolates to reduce esculin, urea and hippurate and to resist lysozyme was also checked. 91.66% of the isolates showed ability to decompose esculin and 63.88% of the isolates had the capacity to produce urease and to decompose urea. | Abstract: | There are a large number of agronomic-ecological interactions that occur in a world with increasing levels of CO2, higher temperatures and a more variable climate. Climate change and the associated severe problems will alter soil microbial populations and diversity. Soils supply many atmospheric green house gases by performing as sources or sinks. The most important of these gases include CH4, CO2 and N2O. Most of the green house gases production and consumption processes in soil are probably due to microorganisms. There is strong inquisitiveness to store carbon (C) in soils to balance global climate change. Microorganisms are vital to C sequestration by mediating putrefaction and controlling the paneling of plant residue-C between CO2 respiration losses or storage in semi-permanent soil-C pools. Microbial population groups and utility can be manipulated or distorted in the course of disturbance and C inputs to either support or edge the retention of C. Fungi play a significant role in decomposition and appear to produce organic matter that is more recalcitrant and favor long-term C storage and thus are key functional group to focus on in developing C sequestration systems. Plant residue chemistry can influence microbial communities and C loss or flow into soil C pools. | Abstract: | Industrialization of our society has led to an increased production and discharge of both xenobiotic and natural chemical substances. Many of these chemicals will end up in the soil. Pollution of soils with heavy metals is becoming one of the most severe ecological and human health hazards. Elevated levels of heavy metals decrease soil microbial activity and bacteria need to develop different mechanisms to confer resistances to these heavy metals. Bacteria develop heavy-metal resistance mostly for their survivals, especially a significant portion of the resistant phenomena was found in the environmental strains. Therefore, in the present work, we check the multiple metal tolerance patterns of bacterial strains isolated from the soils of MG University campus, Kottayam. A total of 46 bacterial strains were isolated from different locations of the campus and tested for their resistant to 5 common metals in use (lead, zinc, copper, cadmium and nickel) by agar dilution method. The results of the present work revealed that there was a spatial variation of bacterial metal resistance in the soils of MG University campus, this may be due to the difference in metal contamination in different sampling location. All of the isolates showed resistance to one or more heavy metals selected. Tolerance to lead was relatively high followed by zinc, nickel, copper and cadmium. About 33% of the isolates showed very high tolerance (>4000μg/ml) to lead. Tolerance to cadmium (65%) was rather low (<100 μg/ml). Resistance to zinc was in between 100μg/ml - 1000μg/ml and the majority of them shows resistance in between 200μg/ml - 500μg/ml. Nickel resistance was in between 100μg/ml - 1000μg/ml and a good number of them shows resistance in between 300μg/ml - 400μg/ml. Resistance to copper was in between <100μg/ml - 500μg/ml and most of them showed resistance in between 300μg/ml - 400μg/ml. From the results of this study, it was concluded that heavy metal-resistant bacteria are widely distributed in the soils of MG university campus and the tolerance of heavy metals varied among bacteria and between locations |

| Abstract: | Soil microorganisms play a main part in organic matter decomposition and are consequently necessary to soil ecosystem processes maintaining primary productivity of plants. In light of current concerns about the impact of cultivation and climate change on biodiversity and ecosystem performance, it is vital to expand a complete understanding of the microbial community ecology in our soils. In the present study we measured the depth wise profile of microbial load in relation with important soil physicochemical characteristics (soil temperature, soil pH, moisture content, organic carbon and available NPK) of the soil samples collected from Mahatma Gandhi University Campus, Kottayam (midland region of Kerala). Soil cores (30 cm deep) were taken and the cores were separated into three 10-cm depths to examine depth wise distribution. In the present study, bacterial load ranged from 141×105 to 271×105 CFU/g (10cm depth), from 80×105 to 131×105 CFU/g (20cm depth) and from 260×104 to 47×105 CFU/g (30cm depth). Fungal load varies from 124×103 to 27×104 CFU/g, from 61×103 to110×103 CFU/g and from 16×103 to 49×103 CFU/g at 10, 20 and 30 cm respectively. Actinomycetes count ranged from 129×103 to 60×104 CFU/g (10cm), from 70×103 to 31×104 CFU/g (20cm) and from 14×103 to 66×103 CFU/g (30cm). The study revealed that there was a significant difference in the depthwise distribution of microbial load and soil physico-chemical properties. Bacterial, fungal and actinomycetes load showed a decreasing trend with increasing depth at all the sites. Except pH all other physicochemical properties showed decreasing trend with increasing depth. The vertical profile of total microbial load was well matched with the depthwise profiles of soil nutrients and organic carbon that is microbial load was highest at the soil surface where organics and nutrients were highest |

| Abstract: | Here we investigate the diversity of pathogenic Vibrio species in marine environments close to Suva, Fiji. We use four distinct yet complementary analyses – biochemical testing, phylogenetic analyses, metagenomic analyses and molecular typing – to provide some preliminary insights into the diversity of vibrios in this region. Taken together our analyses confirmed the presence of nine Vibrio species, including three of the most important disease-causing vibrios (i.e. V. cholerae, V. parahaemolyticus and V. vulnificus), in Fijian marine environments. Furthermore, since toxigenic V. parahaemolyticus are present on fish for consumption we suggest these bacteria represent a potential public health risk. Our results from Illumina short read sequencing are encouraging in the context of microbial profiling and biomonitoring. They suggest this approach may offer an efficient and costeffective method for studying the dynamics of microbial diversity in marine environments over time. |

| Abstract: | Emergence of drug resistance among pathogenic bacteria to currently available antibiotics has intensified the search for novel bioactive compounds from unexplored habitats. In the present study actinomycetes were isolated from two relatively unexplored and widely differing habitats such as mountain and wetlands and their ability to produce antibacterial substances were analyzed. Pure cultures of actinomycetes were identified by morphological and biochemical tests. Various genera of actinomycetes encountered included Nocardia, Pseudonocardia, Streptomyces, Nocardiopsis, Streptosporangium, Micromonospora, Rhodococcus, Actinosynnema, Nocardiodes, Kitasatosporia, Gordona, Intrasporangium and Streptoalloteichus. The frequency of occurrence of each genus was found to vary with sample. About 47% of wetland isolates and 33% of mountain isolates were identified as various species of Nocardia. The isolated strains differed among themselves in their ability to decompose proteins and amino acids and also in enzyme production potential. Antibiotic activities of these actinomycetes were evaluated against 12 test pathogenic bacteria by well diffusion method using agar wells in glycerol-yeast extract agar. About 95% of actinomycete isolates from wetland ecosystem and 75% of highland isolates suppressed in different degrees the growth of test pathogens. Relatively high antibacterial activity among these isolates underlined their potential as a source of novel antibiotics. |

| Abstract: | Present study is focused on the spatiotemporal variation of the microbial population (bacteria, fungus and actinomycetes) in the grassland soils of tropical montane forest and its relation with important soil physico-chemical characteristics and nutrients. Different physico-chemical properties of the soil such as temperature, moisture content, organic carbon, available nitrogen, available phosphorous and available potassium have been studied. Results of the present study revealed that both microbial load and soil characteristics showed spatiotemporal variation. Microbial population of the grassland soils were characterized by high load of bacteria followed by fungus and actinomycetes. Microbial load was high during pre monsoon season, followed by post monsoon and monsoon. The microbial load varied with important soil physico-chemical properties and nutrients. Organic carbon content, available nitrogen and available phosphorous were positively correlated with bacterial load and the correlation is significant at 0.05 and 0.01 levels respectively. Available nitrogen and available phosphorous were positively correlated with fungus at 0.05 level significance. | Abstract: | This study aimed at detecting the prevalence of antibiotic-resistant serotypes of Escherichia coli in Cochin estuary, India. E. coli strains were isolated during the period January 2010–December 2011 from five different stations set at Cochin estuary. Water samples from five different stations in Cochin estuary were collected on a monthly basis for a period of two years. Isolates were serotyped, antibiogram-phenotyped for twelve antimicrobial agents, and genotyped by polymerase chain reaction for uid gene that codes for β-D-glucuronidase. These E. coli strains from Cochin estuary were tested against twelve antibiotics to determine the prevalence of multiple antibiotic resistance among them. The results revealed that more than 53.33% of the isolates were multiple antibiotic resistant. Thirteen isolates showed resistance to sulphonamides and two of them contained the sul 1 gene. Class 1 integrons were detected in two E. coli strains which were resistant to more than seven antibiotics. In the present study, O serotyping, antibiotic sensitivity, and polymerase chain reaction were employed with the purpose of establishing the present distribution of multiple antibiotic-resistant serotypes, associated with E. coli isolated from different parts of Cochin estuary. |

| Abstract: | The production of heavy metals has increased quickly since the industrial revolution. Heavy metals frequently form compounds that can be toxic, carcinogenic, or mutagenic, even in very small concentrations. The usual techniques of removing metals from wastewaters are in general expensive and have many restrictions. Alternative methods of metal removal and recovery based on biological materials have been measured. Among various agents, the use of microbes for the removal of metals from industrial and municipal wastewater has been proposed as a promising alternative to conventional heavy metal management strategies in past decades. Thus, the present study aims to isolate and characterize bacteria from soil, sediment, and waters of metal-contaminated industrial area to study the zinc resistance patterns and the zinc bioaccumulation potential of the selected microorganism. Zinc analysis of the samples revealed that concentrations varying from 39.832 m g/L to 310.24 m g/L in water, 12.81 m g/g to 407.53 m g/g in soil, and 81.06 m g/g to 829.54 m g/g in sediment are present. Bacterial zinc resistance study showed that tolerance to Zn was relatively low (<500 m g/ml). Ten bacterial genera were represented in soil and 11 from water, while only 5 bacterial genera were recorded from sediment samples. Bacillus, Pseudomonas , and Enterobacter were found in soil, sediment, and water samples. Highly zincresistant Bacillus sp. was selected for zinc removal experiment. Zinc removal studies revealed that at pH 5 about 40% reduction occurs; at pH 7, 25% occurs; and at pH 9, 50% occurs. Relatively an increased removal of Zinc was observed in the fi rst day of the experiment by Bacillus sp. The metal bioaccumulative potential of the selected isolates may have possible applications in the removal and recovery of zinc from industrial ef fluents. |

| Abstract: | Water quality of rooftop-collected rainwater is an issue of increased interest particularly in developing countries where the collected water is used as a source of drinking water. Bacteriological and chemical parameters of 25 samples of rooftop-harvested rainwater stored in ferrocement tanks were analyzed in the study described in this article. Except for the pH and lower dissolved oxygen levels, all other physicochemical parameters were within World Health Organization guidelines. Bacteriological results revealed that the rooftop-harvested rainwater stored in tanks does not often meet the bacteriological quality standards prescribed for drinking water. Fifty percent of samples of harvested rainwater for rural and urban community use and 20% of the samples for individual household use showed the presence of E. coli. Fecal coliform/fecal streptococci ratios revealed nonhuman animal sources of fecal pollution. Risk assessment of bacterial isolates from the harvested rainwater showed high resistance to ampicillin, erythromycin, penicillin, and vancomycin. Multiple antibiotic resistance (MAR) indexing of the isolates and elucidation of the resistance patterns revealed that 73% of the isolates exhibited MAR |

| Abstract: | Contamination of environmental water by pathogenic microorganisms and subsequent infections originated from such sources during different contact and non- contact recreational activities are a major public health problem worldwide particularly in developing countries. The main pathogen frequently associated with enteric infection in developing countries are Salmonella enterica serovar typhi and paratyphi. Although the natural habitat of Salmonella is the gastrointestinal tract of animals, it find its way into natural water through faecal contamination and are frequently identified from various aquatic environments (Baudart et al., 2000; Dionisio et al., 2000; Martinez -Urtaza et al., 2004., Abhirosh et al., 2008). Typhoid fever caused by S. enterica serotype typhi and paratyphi are a common infectious disease occurring in all the parts of the world with its highest endemicity in certain parts of Asia, Africa, Latin America and in the Indian subcontinent with an estimated incidence of 33 million cases each year with significant morbidity and mortality (Threlfall, 2002). In most cases the disease is transmitted by polluted water (Girard et al., 2006) because of the poor hygienic conditions, inadequate clean water supplies and sewage treatment facilities. | Abstract: | The objective of the study was to evaluate the survival response of multi-drug resistant enteropathogenic Escherichia coli and Salmonella paratyphi to the salinity fluctuations induced by a saltwater barrier constructed in Vembanadu lake, which separates the lake into a freshwater dominated southern and brackish water dominated northern part. Therefore, microcosms containing freshwater, brackish water and microcosms with different saline concentrations (5, 10, 15, 20, 25 ppt) inoculated with E. coli/S. paratyphi were monitored up to 34 days at 20 and 30 WC. E. coli and S. paratyphi exhibited significantly higher (p <0.05) survival at 20 WC compared to 30 WC in all microcosms. Despite fresh/brackish water, E. coli and S. paratyphi showed prolonged survival up to 34 days at both temperatures. They also demonstrated better survival potential at all tested saline concentrations except 25 ppt where a significantly higher (p<0.0001) decay was observed. Therefore, enhanced survival exhibited by the multi-drug resistant enteropathogenic E. coli and S. paratyphi over a wide range of salinity levels suggest that they are able to remain viable for a very long time at higher densities in all seasons of the year in Vembanadu lake irrespective of saline concentrations, and may pose potential public health risks during recreational activities |
